Description
Tandem mass tag (TMT) isobaric label reagents are used for multiplexed protein quantitation using tandem mass spectrometry. Immobilized Anti-TMT Resin is designed to effectively capture and enrich peptides or proteins labeled using any TMT reagent. Anti-TMT Resin and TMT Elution Buffer are effective for reducing sample complexity, improving dynamic range, and studying cysteine modifications such as S-nitrosylation, oxidation, and disulfide bonds of protein samples labeled with iodoTMT reagents.
Tandem mass tag (TMT) reagents are used to chemically label proteins in different samples for multiplexed quantitation using tandem mass spectrometry. Immobilized Anti-TMT Resin contains an agarose-immobilized anti-TMT monoclonal antibody and is effective for capture and enrichment of peptides or proteins labeled using any TMT reagent. This helps simplify otherwise complex samples before LC-MS/MS analysis, especially when the targets for labeling are rare (e.g., cysteine sulfhydryls). Enrichment of labeled peptides by this method results in fewer peptides identified per protein, but can significantly increase the number of unique proteins identified, making it easier to detect and measure low-abundance proteins.
Immobilized Anti-TMT Resin uses our highly specific Anti-TMT Antibody (Cat. No. 90075) to bind peptides labeled with TMT reagents. The antibody is specific for the mass reporter region of the TMT reagents, which allows for enrichment of TMT-labeled peptides regardless of peptide labeling chemistry (e.g., amine-reactive, sulfhydryl-reactive). TMT Elution Buffer is a volatile neutral buffer that competitively elutes captured TMT-labeled peptides. When used in combination with Immobilized Anti-TMT Resin, TMT Elution Buffer provides higher specific enrichment of TMT-labeled peptides compared to acidic or basic elution buffers.
