The Human Syndecan 1 (SDC1) ELISA quantitates Hu Syndecan 1 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu Syndecan 1. Principle of the method The Human Syndecan 1 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. CD138 (Syndecan 1) is a transmembrane proteoglycan made up of one core protein and five glycosaminoglycans that can bind a variety of cytokines and modulate their activity, as well as the activity of extracellular matrix components and influence many developmental processes. CD138 is expressed mainly in differentiating keratinocytes and is transiently upregulated in all layers of the epidermis upon tissue injury. CD138 is also highly expressed on plasma cells and can be detected even on fibroblasts, vascular smooth muscle cells and endothelial cells. Up-regulation and down-regulation of CD138 on the cell surface often correlates with the gain of cancerous characteristics. Serum levels of the shedded soluble sCD138 are used as a prognostic factor of carcinogenesis. CD138 is expected to play a role in cell adhesion, cell proliferation, cell migration and cell-matrix interactions via its receptor for extracellular matrix proteins. Further, CD138 is expressed on the surface of pre-B cells and plasma cells but is absent from mature B cells, and is lost from the apoptotic myeloma cells. Altered CD138 expression has been detected in several different tumor types. While several transcript variants may exist for the CD138 gene, the full-length natures of only two have been described to date which represent the major variants and encode the same protein.Specifications
