Description
- Recognizes human PIGF
- PIGF (Placenta growth factor, PGF) is an angiogenic factor belonging to vascular endothelial growth factor family
- Located on chromosome 14q14, encodes four isoforms of PIGF
- Protein secreted as glycosylated homodimer
- PIGF-1 and 3 are diffusible isoforms, PIGF-2 and 4 have heparin binding domains
- PIGF identified in placenta, regulates growth and differentiation of trophoblasts, expressed in umbilical vein endothelial cells and non-placental tissues (thyroid gland and developing lung tissue)
- Has angiogenic properties, enhancing survival, growth and migration of endothelial cells in vitro and promotes vessel formation in certain in vivo models
- Potential biomarker for preeclampsia
Process:
- Antibody specific for PIGF pre-coated onto microwells
- PIGF protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for PIGF is added to detect captured PIGF protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
